Cell Line STR Profiling Analysis Services

Creative Bioarray is able to implement cell line certification STR analysis services to help customers investigate possible intra-species contamination and inter-species cross-contamination. The results of this service can be used to assist journal publication, or to troubleshoot possible human errors in experiments. We have established a mature protocol for STR genotyping to provide you with a one-stop service.

Cell Line Authentication and STR

According to the recommendations of the American Type Culture Collection (ATCC) Standards Development Organization Working Group ASN-0002, short tandem repeat (STR) analysis is a standard test for validating cell lines. The analysis of highly variable STRs provides a simple, inexpensive, and specific genetic "fingerprint" of cell lines. Cell line STR analysis confirms cell identity by analyzing DNA microsatellite regions with the variable numbers of repeats and located in the entire genome. The important thing is that STR analysis is the test required by most journals. At present, STR markers for human cell lines have formed a relatively complete genotype profile of cell lines, but the standardization method for STR purification of non-human cells needs to be further improved. Our STR analysis service is mainly carried out for human cell lines, and also accepts STR analysis of cell lines of experimental animals such as mice and rats.

Genotyping for Cell Line Authentication Services

Service Purposes

Intraspecies authentication of human cell lines: to verify whether the cell line in question is of human origin by detecting the absence of standard markers;

Interspecies cross-contamination analysis service: Species-specific authentication can detect cross-contamination between common animal types such as pigs, cats, Chinese hamsters, rhesus monkeys, sheep, horses, rats, dogs and cattle, and human cell lines;

Selection of STR Markers

According to the basic source information (species, cell line names, and potentially contaminating cell names) of the cell samples submitted by customers, our experts will use NCBI's BLAST tool to design STR markers. We provide detection of at least 8 STR loci, and customers can choose to increase the number of labels according to the situation.

Primer Design

Design multiple PCR primers according to the STR tags planned in the designed scheme.

STR Analysis

We use the internally designed multiplex PCR standardization process to perform PCR amplification on the samples, and obtain the amplicon sequence through multiplex PCR. The resulting amplicons are separated using capillary electrophoresis to determine the number of replicates in the sample.

Data Analysis

The STR data of our service can be compared with the STR profiles of some STR databases (ATCC, DSMZ, JCRB, RIKEN, CLIMA, MD Anderson, Sanger). The purpose of this is to verify whether the genotype of the cell line in question is the same as the genotype of the putative parental cell.
