Detailed Separation Procedure
The isolation of CD8+CD45RA+ effector T cells is performed in a two-step procedure. First, non-target cells are indirectly magnetically labeled for depletion with biotin-conjugated antibodies against CD45RO and CD56 and an APC-conjugated antibody against CD197 (CCR7), as well as Anti-APC Microbeads and Anti-Biotin Microbeads. In the second step, the CD8+CD45RA+ effector T cells are directly labeled with CD8 Microbeads and isolated by positive selection from the pre-enriched cell fraction.
