Description

THAP11 (THAP domain-contining protein 11), also known as Ronin, is an essential factor involved in embryonic stem (ES) cell pluripotency and cell growth. THAP11 is regulated by shuttling between cytoplasm-nucleus, and is considered a transcriptional repressor that plays a central role in embryogenesis as well as in ES cell pluripotency. It acts as a sequence-specific DNA-binding factor which represses gene expression in pluripotent ES cells by directly binding to key genetic loci and recruiting epigenetic modifiers. RONIN is the mouse homolog of THAP11 and its homozygous deletion happened to be embryonically lethal in mice wherein was shown to be essential factor underlying embryogenesis/ES cell pluripotency via its activational/repressional effects on transcription. THAP11 also functions as a negative regulator of cell growth in human HepG2 hepatoma cells through transcriptional repression of the proto-oncogene MYC.
